Sally Sue and Western Blots

You're probably wondering exactly how Sally Sue does Simple Westerns.

Well, everything happens inside of a capillary.

Sally Sue picks up 12 capillaries at a time for each cycle.

She does up to 8 cycles for a total of 96 Simple Westerns.

Sally Sue will load samples, antibodies, and reagents into each capillary from a 384-well plate.

She begins by first loading the separation matrix and then a stocking matrix into each capillary.

Sally Sue then loads your protein lysate.

The capillaries are placed into the separation tray where a voltage is applied across each capillary to enable separation by molecular weight.

Once the separation is complete, the capillaries are exposed to ultraviolet (UV) light which immobilizes your proteins to the capillary walls.

This allows Sally Sue to perform an immunoassay within the capillary by first incubating with primary antibody and then a Horseradish Peroxidase (HRP) conjugated secondary antibody followed by a chemiluminescence substrate.

This chemiluminescence reaction gives off light that is read by a CCD camera.

Your quantitative results are automatically generated for you and reported by electropherogram or lane view.
